Balancing act: matching growth with environment by the TOR signalling pathway.

چکیده
One of the most fundamental aspects of growth in plants is its plasticity in relation to fluctuating environmental conditions. Growth of meristematic cells relies predominantly on protein synthesis, one of the most energy-consuming activities in cells, and thus is tightly regulated in accordance with the available nutrient and energy supplies. The Target of Rapamycin (TOR) signalling pathway takes a central position in this regulation. The core of the TOR signalling pathway is conserved throughout evolution, and can be traced back to the last eukaryotic common ancestor. In plants, a single complex constitutes the TOR signalling pathway. Manipulating the components of the TOR complex in Arabidopsis highlighted its common role as a major regulator of protein synthesis and metabolism, that is also involved in other biological functions such as cell-wall integrity, regulation of cell proliferation, and cell size. TOR, as an integral part of the auxin signalling pathway, connects hormonal and nutrient pathways. Downstream of TOR, S6 kinase and the ribosomal S6 protein have been shown to mediate several of these responses, although there is evidence of other complex non-linear TOR signalling pathway structures.

I-7: Maternal Signalling to the Placenta
Background: Though it is well established that maternal blood-borne signals influence highly the growth of the placenta, the mechanisms are not known. In vitro trophoblast culture models are limited by an inability to reconstruct the polarised bilayer of the human hemochorial placenta. We have used a first trimester villous tissue explant system to investigate how growth factors interact with p...
متن کاملThe nutrient transceptor/PKA pathway functions independently of TOR and responds to leucine and Gcn2 in a TOR-independent manner
Two nutrient-controlled signalling pathways, the PKA and TOR pathway, play a major role in nutrient regulation of growth as well as growth-correlated properties in yeast. The relationship between the two pathways is not well understood. We have used Gap1 and Pho84 transceptor-mediated activation of trehalase and phosphorylation of fragmented Sch9 as a read-out for rapid nutrient activation of P...
متن کاملThe Effect of Ampk and P53 Proteins On Tor Pathway Following Endurance Training In The Left Ventricle Of The Heart Of Diabetic Rats By Streptozotocin And Nicotinamide
Background: AMPK and P53 proteins regulate the TOR protein in the TORC1 complex, which regulates many physiological processes. The aim of this study was to evaluate the effect of AMPK and P53 proteins on the TOR pathway following endurance training in the left ventricle of the heart of diabetic rats by streptozotocin and nicotinamide. Methods: In this experimental study, 12 head two-month-old ...
متن کاملThe TOR nutrient signalling pathway phosphorylates NPR1 and inhibits turnover of the tryptophan permease.
The Saccharomyces cerevisiae targets of rapamycin, TOR1 and TOR2, signal activation of cell growth in response to nutrient availability. Loss of TOR or rapamycin treatment causes yeast cells to arrest growth in early G1 and to express several other physiological properties of starved (G0) cells. As part of this starvation response, high affinity amino acid permeases such as the tryptophan perme...
متن کاملNutrient/TOR-dependent regulation of RNA polymerase III controls tissue and organismal growth in Drosophila.
The nutrient/target-of-rapamycin (TOR) pathway has emerged as a key regulator of tissue and organismal growth in metazoans. The signalling components of the nutrient/TOR pathway are well defined; however, the downstream effectors are less understood. Here, we show that the control of RNA polymerase (Pol) III-dependent transcription is an essential target of TOR in Drosophila. We find that TOR a...
